Comparison of Gene Expression and Genome-Wide DNA Methylation Profiling between Phenotypically Normal Cloned Pigs and Conventionally Bred Controls

ABSTRACT
Animal breeding via Somatic Cell Nuclear Transfer (SCNT) has enormous potential in agriculture and biomedicine. However, concerns about whether SCNT animals are as healthy or epigenetically normal conventionally bred ones raised the efficiency of cloning by is much lower than natural In-vitro fertilization (IVF). Thus, we have conducted a genome-wide gene expression DNA methylation profiling between phenotypically cloned pigs control two tissues (muscle liver), using Affymetrix Porcine array well modified methylation-specific digital karyotyping (MMSDK) Solexa sequencing technology. Typical tissue-specific differences with respect to both were observed muscle liver from pigs. Gene profiles highly similar controls, though small set genes showed altered expression. Cloned presented more different pattern unique sequences tissues. Especially genomic sites had status trend towards slightly increased levels Molecular network analysis that contained such differential loci revealed significant related tissue development. In conclusion, our study their expression, but moderate alteration aspects still exists, especially certain regions.
