ABSTRACT
Exposure of Lead (Pb), a known neurotoxicant, can impair spatial learning and memory probably via impairing the hippocampal long-term potentiation (LTP) as well neuronal injury. Activation microglia also impairs memory. Thus, we raised hypothesis that activation is involved in Pb exposure induced LTP impairment To test this clarify its underlying mechanisms, investigated Pb-exposure on activation, cytokine release, level injury vivo or vitro model. The changes these parameters were observed after pretreatment with minocycline, inhibitor. Long-term low dose (100 ppm for 8 weeks) caused significant reduction acute slice preparations, meanwhile, such treatment significantly increased In increase up-regulate release cytokines including tumor necrosis factor-alpha (TNF-α), interleukin-1β (IL-1β) inducible nitric oxide synthase (iNOS) culture alone co-culture neurons. Inhibiting minocycline reversed above-mentioned changes. Our results showed cause which IL-1β, TNF-α iNOS, proinflammatory factors may deficits.
