ABSTRACT
Hyperglycemia, the diagnostic feature of diabetes also occurs in non-diabetics associated with chronic inflammation and systemic insulin resistance. Since increased risk active TB diabetics has been linked to severity duration hyperglycemia, we investigated what effect diet-induced hyperglycemia had on Mycobacterium tuberculosis (Mtb) infection non-diabetic guinea pigs. Post-prandial was induced pigs normal chow by feeding a 40% sucrose solution daily or water as carrier control. Sucrose initiated day aerosol exposure H37Rv strain Mtb continued for 30 60 days infection. Despite more severe sucrose-fed animals 30, there no significant difference lung bacterial lesion burden until 60. However higher spleen lymph node at indicated earlier extrapulmonary animals. In both sucrose- water-fed animals, serum free fatty acids, important mediators resistance, were remained elevated Hyperglycemia mediated resulted accumulation advanced glycation end products (AGEs) granulomas, which exacerbated feeding. However, tissue AGEs These data indicate that alone induces resistance is Moreover, AGEs, are central pathogenesis diabetic complications. The exacerbation may explain why poorly controlled compared patients properly blood glucose levels.
