ABSTRACT
Aims of the Study To investigate neuroprotective effect Lycium barbarum polysaccharide (LBP) on focal cerebral ischemic injury in mice and to explore its possible mechanism. Materials Methods Male ICR were used make model middle artery occlusion (MCAO) after intragastric administration with LBP (10, 20 40 mg/kg) Nimodipine (0.4 for seven successive days. After 24 h reperfusion, neurological scores estimated infarct volumes measured by 2, 3, 5-triphenyltetrazolium chloride (TTC) staining. Morphological changes brains performed hematoxylin-eosin (HE) The number apoptotic neurons was detected TUNEL Bax, Bcl-2 protein expression CytC, Caspase-3, -9 cleaved PARP-1 activation investigated immunofluorescence western-blot analysis. Results treatment groups significantly reduced infract volume deficit scores. also relieved neuronal morphological damage attenuated apoptosis. at dose mg/kg suppressed overexpression PARP-1, inhibited reduction expression. Conclusions Based these findings we propose that protects against attenuating mitochondrial apoptosis pathway.
