ABSTRACT
The identification of suitable reference genes is critical for obtaining reliable results from gene expression studies using quantitative real-time PCR (qPCR) because the may vary considerably under different experimental conditions. In most cases, however, commonly used are employed in data normalization without proper validation, which lead to incorrect interpretation. Here, we aim select a set optimal accurate associated with intramuscular fat (IMF) deposition during development. present study, eight (PPIB, HMBS, RPLP0, B2M, YWHAZ, 18S, GAPDH and ACTB) were evaluated by three algorithms (geNorm, NormFinder BestKeeper) two types muscle tissues (longissimus dorsi biceps femoris muscle) across developmental stages. All gave similar results. PPIB HMBS identified as stable genes, while 18S variably expressed, varying dramatically Furthermore, reveal crucial role appropriate result, analysis PPARG was performed least genes. relative levels normalized greatly differed those one. Therefore, evaluation must be given condition before used. IMF skeletal
