ABSTRACT
Background We evaluated the side effects of bisphosphonate (BP) on tooth extraction socket healing in spontaneously diabetic Torii (SDT) rats, an established model non-obese type 2 diabetes mellitus, to develop animal BP-related osteonecrosis jaws (BRONJ). Materials and Methods Male Sprague-Dawley (SD) rats SDT were randomly assigned zoledronic acid (ZOL)-treated groups (SD/ZOL or SDT/ZOL) control (SD/control SDT/control). Rats SD/ZOL SDT/ZOL received intravenous bolus injection ZOL (35 μg/kg) every weeks. Each group consisted 6 each. Twenty-one weeks after treatment began, left maxillary molars extracted. The euthanized at 2, 4, 8 extraction, total maxillae harvested for histological histochemical studies. Results In oral cavity, bone exposure persisted site all until extraction. contrast, there was no SD/control SDT/control groups, only 1 showed exposure. Histologically, necrotic areas with empty lacunae, microbial colonies, less invasion by inflammatory cells observed. number tartrate-resistant phosphatase-positive osteoclasts lower than group. mineral apposition rate significantly compared Conclusions This study demonstrated development BRONJ-like lesions suggested that low turnover cell infiltration plays important role BRONJ.
