ABSTRACT
Diagnostic tests for veterinary surveillance programs should be efficient, easy to use and, possibly, economical. In this context, classic Enzyme linked ImmunoSorbent Assay (ELISA) remains the most common analytical platform employed serological analyses. The analysis of pooled samples instead individual ones is a procedure that permits certify, with one single test, entire herds as "disease-free". However, diagnostic need particularly sensitive, especially when levels disease markers are low, in case anti-BoHV1 antibodies milk Infectious Bovine Rhinotracheitis (IBR) disease. avidin-nucleic-acid-nanoassembly (ANANAS) novel kind signal amplification immunodiagnostics based on colloidal poly-avidin nanoparticles that, using model analytes, was shown strongly increase ELISA test performance compared monomeric avidin. Here, first time, we applied ANANAS reagent integration real context. monoclonal 1G10 anti-bovine IgG1 antibody biotinylated and integrated reagents indirect IBR diagnosis from mimicking tank prevalence between 1 8%. sensitivity specificity method same directly horseradish peroxidase, commercial IDEXX kit recently introduced market. increased by 5-fold mAb-based conventional without loosing specificity. When kit, 1G10-ANANAS capable detect presence anti-BHV1 bulk gE positive animals 2-fold higher similar results demonstrate potentials new technology, which improving current limits hardware investments.
