ABSTRACT
Mast cells (MCs) are a versatile cell type playing key roles in tissue morphogenesis and host defence against bacteria parasites. Furthermore, they can enhance immunological danger signals implicated inflammatory disorders like fibrosis. This granulated originates from the myeloid lineage has similarities to basophilic granulocytes, both containing large quantities of histamine heparin. Immature murine mast mature their destination adopt either connective (CTMC) or mucosal (MMC) type. Some effector functions executed by activation/degranulation MCs which lead secretion typical set MC proteases (MCPT) preformed newly synthesized mediators its granules into local microenvironment. Due potential accumulation mutations signalling pathway components corresponding cell-lines, primary cultured an attractive mean study general features biology aspects relevant human disease. Here, we describe simple protocol for simultaneous isolation CTMC-like peritoneum (PMCs) immature precursors bone marrow (BM). The latter differentiated vitro yield BM-derived (BMMC). These display morphological phenotypic MCs, express surface markers, be propagated kept culture several weeks. provided allows amplification homogenous, non-transformed marrow-derived cell- tissue-based biomedical research.
