ABSTRACT
Thrombospondin 1 and thrombospondin 2 (THBS1 THBS2) share similar multifunctional domains, are known to be antiangiogenic. However, the expression pattern of THBS1 THBS2 is different, specific role in different subtypes lung cancer remains largely unclear. To evaluate significance development cancer, present study performed a microarray-based systematic-analysis determine transcript levels thrombospondins their relation prognosis cancer. was general underexpressed cancer; contrast, mRNA were markedly overexpressed number datasets non-small cell carcinoma (NSCLC), including adenocarcinoma (AC) squamous carcinoma. Similar verified pulmonary AC lines with real-time PCR analysis. The survival patients high poorer than low THBS2. In analysis, genes coexpressed or determined. Pulmonary level sevenTSHB1-coexpressed (CCL5, CDH11, FYB, GZMK, LA-DQA1, PDE4DIP, SELL) had better rates those level. Patients seven TSHB2-coexpressed (CHI3L1, COL5A2, COL11A1, FAP, MXRA5, THY1, VCAN) poor rates. Downregulation VCAN shRNA inhibited proliferation A549 line. summary, functions as tumor suppressor adenocarcinoma. may play double-edged progression AC, i.e. anti-angiogenic oncogenic function. Further on mechanism underlying activity warranted have further implications for diagnosis treatment AC.
