ABSTRACT
The analysis of intracellular ATP can reveal the response cells to different treatments and is important for individualized medicine. In present study, we developed a cell penetrating peptides (CPPs) tagged luciferase (TAT-LUC) tumor chemosensitivity assay. activity recombinant TAT-LUC was evaluated using standard solution cells. This then used sensitivity index (SI) four strains results showed that could detect less than 10 nM extracellular with strong correlation between luminescence intensity content (R2 = 0.994). Without lysis, detection limit 40 Furthermore, (Skov-3/DDP, A549/DDP, MDA-MB-231, Huh-7) determined by this assay successfully. penetration ability enables not only reflect drug resistance real-timely but also minimize test time, which be valuable aid personalized cancer chemotherapy.
