ABSTRACT
Pathogenic or non-pathogenic small (17 to 30 nt) and long (>200 non-coding RNAs (ncRNAs) have been implicated in the regulation of gene expression at transcriptional, post-transcriptional epigenetic level by interacting with host proteins. However, lack suitable experimental system precludes identification evaluation functional significance proteins ncRNAs. In this study, we present a first report on application riboproteomics identify small, highly pathogenic, noncoding satellite RNA (sat-RNA) associated Cucumber mosaic virus, helper virus (HV). affinity beads containing sat-RNA transcripts (+) (-)-sense covalently coupled cyanogen bromide activated sepharose were incubated total protein extracts from either healthy HV-infected Nicotiana benthamiana leaves. RNA-protein complexes bound eluted subjected MudPIT analysis. Bioinformatics programs PANTHER classification WoLF-PSORT used further classify identified each case based their functionality subcellular distribution. Finally, observed that network plus minus-strand sat-RNA, presence absence HV is distinct, global interactome satRNA orientations very different.
