ABSTRACT
The effect of apatinib on the formation vasculogenic mimicry (VM) was studied in a malignant melanoma cell line. MUM-2B cells cultured three-dimensional Matrigel were treated with varying concentrations (0, 0.01, 0.05, 0.1, 0.5 μmol/L) to test its VM vitro, followed by MTT proliferation and transwell invasion assays determine cells. In vivo, we used cancer model short-term (100, 200, 300 mg/kg) treatment VM. Western blotting, immunohistochemistry staining, CD31-PAS dual staining performed assess expression VEGFR-2, ERK-1/2, PI3K, MMP-2, results showed apatinib-treated groups formed lesser number 3D matrigel, while viability assay migration significantly lower groups. addition, inhibited angiogenesis, formation, tumor growth models cancer. Mice markedly reduced MMP-2. summary, could inhibit downregulate ERK1/2/PI3K/MMP-2 signaling cascade, which may be one underlying mechanisms inhibits angiogenesis development cancer, restrains Apatinib shows inhibitory effects cells, is close relationship
