ABSTRACT
Beijing strains of Mycobacterium tuberculosis (lineage 2) have been associated with drug-resistance and transmission worldwide. Most the identified in Colombian Pacific coast exhibited a multidrug resistant (MDR) phenotype. We sought to evaluate clonality sublineage circulating Southwestern Colombia. Thirty-seven were through spoligotyping out 311 clinical isolates collected 9 years from 2002-2010. Further analysis by MIRU-VNTR 24 loci was conducted for strains. For classification, deletions RD105, RD207, RD131 point mutations at fbpB, mutT2, acs evaluated. Drug-resistance first- second-line anti-TB drugs also Additionally, two Illumina-whole genome sequenced (one MDR one drug-susceptible). Among 37 characterized, 36 belonged SIT190 type which 28 MDR, four pre-extensively drug (XDR) TB, XDR-TB. The remaining strain SIT1 susceptible. allowed identification three clusters unique confirmed as "modern" sublineage. rpoB S531L katG S315T most common among Moreover, evaluated whole sequencing (WGS) shared genetic features 2.2.1 previously characterized Asian WGS revealed presence eight SNPs reported other "Beijing-like" Colombia, them phenotype, suggests different origin this M. compared found neighboring South American countries. This work may serve baseline study evolution spread play an important role propagation MDR-TB.
