ABSTRACT
The aim of this work was to study the effect a high sodium (HS) diet on blood pressure and renal function in male adult rats that have been treated with dual Endothelin receptor antagonist (ERA) during their early postnatal period (day 1 20 life). Male Sprague-Dawley were divided four groups: CNS: control normosodic diet; ERANS: ERA-treated CHS: ERAHS: HS diet. Systolic (SBP) recorded before after 24-hour metabolic cage studies performed. AQP2 α-ENac expressions measured by western blot real time PCR medulla. Vasopressin (AVP) pathway evaluated measuring V2 adenylyl cyclase 6 (AC6) expression cAMP production Pre-pro ET-1mRNA also Only had an ERA increased SBP consumption diet, showing impaired capacity excrete water, i.e. developing salt sensitivity. This sensitivity would be mediated increase renomedullary activity α-ENaC as consequence AC6 and/or decreased ET-1 knowledge molecular mechanisms underlying perinatal programming sensitive hypertension will allow development reprogramming strategies order avoid pathology.
