Anti-inflammatory activity of soluble chito-oligosaccharides (CHOS) on VitD3-induced human THP-1 monocytes

ABSTRACT
Chito-oligosaccharides (CHOS) are oligomers of D-glucosamine and N -acetyl-glucosamine. Anti-inflammatory activities a wide variety CHOS mixtures have previously been reported, mainly based on studies with mouse models murine macrophages. Since the human immune systems quite different, gaining insight into activity cell lines, using well-characterized mixtures, is considerable interest. Bacillus subtilis chitosanase (BsCsn46A) can efficiently convert chitosan to water soluble low molecular weight CHOS. Here, anti-inflammatory properly characterized mixture was studied, THP-1 cells that were differentiated mature monocytes vitamin D3. Addition reduced production multiple pro-inflammatory cytokines associated bacterial lipopolyssacharide (LPS)-stimulated inflammation, in dose-dependent manner without affecting viability. Interestingly, only minimal effects observed similar experiments phorbol 12-myristate 13-acetate- (PMA-) differentiated, macrophage-like, cells. Altogether, addition showing promising biological system, present study also points at Vitamin D3-stimulated as favorable system for assessing bioactive compounds.
