ABSTRACT
Background We have developed a new clinical research approach for the quantification of cellular proliferation in human infants to address unanswered questions about tissue renewal and regeneration. The consists oral 15 N-thymidine administration label cells S-phase, followed by Multi-isotope Imaging Mass Spectrometry detection incorporated cell nuclei. To establish approach, we performed an observational study examine uptake elimination N-thymidine. compared at-home with in-hospital tetralogy Fallot, form congenital heart disease, failure. Methods examined urine samples from 18 who received (50 mg/kg body weight) mouth five consecutive days. used Isotope Ratio determine enrichment N relative 14 (%) urine. Results/findings dose produced periodic rises Infants Fallot had 3.2-fold increase failure 4.3-fold mean peak over baseline. was not statistically different between two patient populations (p = 0.103). time 6.3 ± 1 hr 7.5 2 (mean SEM). duration significant after 18.5 1.7 18.2 1.8 were also 0.617 p 0.887). Conclusions presented results support conclusions significance future applications: (1) Demonstration that at home is equivalent administration. (2) Two types disease show no differences absorption elimination. This enables comparative analysis disease.
