ABSTRACT
The mucosal events of HIV transmission have been extensively studied, but the role infected cells present in genital and rectal secretions, semen, particular, remains a matter debate. As prerequisite to thorough vivo investigation early through cells, we characterized detail by multi-parameter flow cytometry changes macaque seminal leukocytes during SIVmac251 infection, focusing on T macrophages dendritic cells. Using immunocytofluorescence targeting SIV proteins real-time quantitative PCR DNA, investigated nature sorted semen from macaques at different stages infection. Finally, cocultured CD4+ with cell line permissive infection assess their infectivity vitro. We found that primary induced strong local inflammation, which was associated an increase number both factors having potential favor cell-associated virus transmission. Semen were productively all infectious Lymphocytes had phenotype expressed activation (CD69 & HLA-DR) migration (CCR5, CXCR4, LFA-1) markers. CD69 expression increased Macrophages predominated CD4, CCR5, MAC-1 LFA-1. Altogether, demonstrated contains two major SIV-target (CD4+ macrophages). Both types can be are endowed markers may facilitate sexual exposure.
