ABSTRACT
Rationale: Pulmonary hypertension encompasses progressive disorders leading to right ventricular dysfunction and early death. Late detection is an important cause of poor clinical outcomes. However, biomarkers that accurately predict the presence pulmonary are currently lacking. Objectives: In this study, we provide evidence blood platelets contain a distinctive ribonucleic acid (RNA) profile may be exploited for hypertension. Methods: Blood platelet RNA was isolated prospectively from 177 prevalent patients with different subtypes as well 195 control subjects clinically not suspected Sequencing libraries were created using SMARTer (Switching Mechanism at 5′ end Template) copy desoxyribonucleic amplification sequenced on Illumina High Throughput platform. RNA-sequencing reads mapped human reference genome, intron-spanning spliced selected. Differential panels calculated by analysis variance statistics. A particle swarm optimization-enhanced classification algorithm built employing development (n = 213 samples) independent validation series 159 samples). Results: We detected total 4,014 RNAs in 177) asymptomatic 195). Gene ontology revealed enhanced concentrations genes related processing, translation, mitochondrial function. optimization-selected panel 408 differentially mediated 93% sensitivity, 62% specificity, 77% accuracy, 0.89 (95% confidence interval, 0.83–0.93) area under curve, negative predictive value 91% series. The prediction score age, sex, smoking, subtype, use hypertension–specific medication or anticoagulants. Conclusions: discriminate subjects. light current diagnostic delays, study starting point further evaluation RNA-based test ultimately improve diagnosis outcomes
