ABSTRACT
In mammals, adiponectin and its receptors (AdipoR1 AdipoR2) mRNAs are expressed in various tissues. However, the cellular expression role of system have never been investigated rat ovary. Here, we report presence adiponectin, AdipoR1 AdipoR2 ovaries, granulosa cells. Using RT-PCR western blot, show that proteins for found ovaries. Immunohistochemistry localized theca-interstitial T-I cells, corpus luteum, oocyte less abundantly KGN human cell line, mRNA protein were undetectable; was weakly expressed, whereas clearly present. Human chorionic gonadotrophin (hCG) injection (48 h) after pregnant mare serum (PMSG) (24 immature rats increased level (protein) by about threefold ( P < 0.05) those (mRNA, 1.5-fold (protein, ovary, levels unchanged. Interestingly, hCG PMSG treatment decreased plasma insulin levels. vitro primary recombinant (5 μg/ml) or absence follicle-stimulating hormone (10 −8 M, 48 had no effect on steroidogenesis. it progesterone secretion twofold oestradiol production 1.6-fold response to insulin-like growth factor-I (IGF-I) M). Furthermore, improved IGF-I-induced IGF-I receptor-β subunit tyrosine phosphorylation ERK1/2 phosphorylation. basal state, also rapidly but transiently ERK1/2, p38 Akt phosphorylations, more lately adenosine 5′-monophosphate-activated kinase (AMPK) Thus, regulated ovary enhances steroidogenesis
