ABSTRACT
Background: Type 2 diabetes (DM2) and chronic kidney disease (CKD) are inflammatory pathologies. Diabetes is characterized by hyperglycemia CKD the gradual irreversible loss of function. Both diseases develop oxidative stress, reactive oxygen species (ROS) play a pivotal role in pathogenesis. This study aimed to determine ROS production granulocytes from renal patients with or without diabetes. Methods: Granulocytes DM2, CKD, CKD-DM2, healthy controls were purified using Ficoll-Hypaque gradient method. Granulocyte generation absence presence PDB (an activator NADPH-oxidase) Concanavalin A (Toll- receptor 3,9 activator) was evaluated luminol-dependent chemiluminescence The cell-free DNA serum CKD-DM2 measured fluorescence method before after hemodialysis. Results: Our results show significant increase compared control (p<0.05). group produced most levels NADPH-oxidase activation. showed ConA. In contrast, mitochondrial (internal) different response. extrusion higher hemodialysis suggesting cell death. Conclusion: demonstrated that high increased It may suggest more severe has worse clinical prognosis.
