Medication-related osteonecrosis of the jaw. Introduction of a new modified experimental model

ABSTRACT
To evaluate a modified experimental model for medication-related osteonecrosis of the jaw (MRONJ) through upper right central incisor extraction followed by intravenous bisphosphonate administration.Forty five rats underwent tooth were divided in 2 groups: Group I - group, 30 received an administration protocol zoledronic acid 35μg/kg into tail vein every two weeks, totalizing four administrations, during eight weeks administration, previously extraction, and II control 15 didn't any medication before extraction. The groups subdivided postoperative periods: 14/28/42 days. Clinical analysis microtomography performed to verify presence osteonecrosis. In addition, descritive histological hematoxylin-eosin stained sections was or necrotic foci.Twelve (40%) rats, from showed clinical signs MRONJ (p=0.005), however, all samples imaginologic findings like osteolysis loss integrity cellular walls (p≤0.001). Microscopic evaluation revealed areas with microbial colonies inflammatory infiltrate animals presented chronology normal wound healing.The after maxillary rats. This new may be considered option study MRONJ.
