ABSTRACT
We reported in an earlier study that using mass spectrometry and bioinformatic analysis demonstrated Rac1 protein might be mostly mitochondrial target the radiosensitization process of nasopharyngeal carcinoma CNE-1 cells. The goal our current was to reveal relationship between Rac1/NADPH pathway cells activator, phorbol 12-myristate 13-acetate (PMA) inhibitor NSC23766. Rac1-GTP expression determined a pulldown assay, location immunofluorescence with laser scanning confocal microscope, NADPH oxidase activity NBT assay reactive oxygen species DCFH-DA probe. apoptosis rate analyzed by flow cytometry, expressions p67(phox) NFκB-p105/p50 were Western blot. After treatment PMA 2 Gy radiation (compared control), activated translocated cell membrane. activity, intracellular concentration increased significantly. NFκB-p50 also increased. However, treated NSC23766 alone or combined irradiation, results just opposite. Overall, these indicate may key involved can increase radiosensitivity cells, signaling process.
