ABSTRACT
Endometrial cancer is the most common of female reproductive system. Combination treatment with specific agents has been widely used as a targeted therapy for cancer. In this study, we aimed to investigate anti-proliferative and apoptotic effects varying concentrations perifosine vitamin D on human endometrial cell line (HEC-1A). HEC-1A cells were exposed (10 μM, 30 μM), (50 nM, 200 nM) combinations both 48 h 72 h. Monitoring proliferation in time-dependent manner was performed xCELLigence RTCA DP The levels BCL2, BAX P53 mRNA expression examined using RT-qPCR. Apoptosis determined Annexin V, which followed by flow cytometry analysis. Ultra-structural morphology analyzed transmission electron microscopy (TEM) perifosine+vitamin combination (30 μM + nM at 10 h) higher than alone. It observed that increased dose-dependent manner. While did not find any significant change levels. TEM examinations cells, appeared lead autophagic death, whereas caused paraptosis-like death non-apoptotic (paraptotic, necrotic) death. Therefore, it considered drugs might be an alternative effective option through activating mechanisms cells.
