ABSTRACT
Si Jun Zi Tang (SJZT) is a classic Traditional Chinese Medicine (TCM) prescription used to treat aging-related diseases. However, the potential molecular mechanisms of anti-aging effects bioactive compounds and their targets remain elusive. In this study, we combined network pharmacology docking with in vivo experiments elucidate mechanism SJZT. A series strategies were predict therapeutic SJZT, including compound screening, pathway enrichment analysis studies. Based on predictions observation outward signs aging, expression levels selected genes proteins possible key subsequently validated analysed using qRT-PCR immunoblotting. Using data mining approach, 235 effective SJZT aging obtained. AKT1, STAT3, JUN, MAPK3, TP53, MAPK1, TNF, RELA, MAPK14 IL6 identified as core Protein-Protein Interaction Networks (PPI) analysis. The results target Gene Ontology (Go) functional suggested that may be involved antiapoptotic biological processes. Kyoto Encyclopedia Genes Genomes (KEGG) indicated associated PI3K-AKT P38 MAPK signalling pathways. Molecular kaempferol quercetin could fit binding pockets targets. addition, alleviated symptoms mice such osteoporosis hair loss. conclusion, effect was inhibition pathways, these findings consistent prediction.
