ABSTRACT
The tumor microenvironment is comprised of cancer cells and various stromal their respective cellular components. Cancer-associated fibroblasts (CAFs), a major part the cells, are key determinant in progression, while glucose-regulated protein (GRP)78 overexpressed many human cancers involved invasion metastasis. This study developed microfluidic-based three dimension (3D) co-culture device to mimic an vitro order investigate cell real-time. bionic chip provided significant information regarding role GRP78, which may be stimulated by CAFs, promote non-small lung vitro. data showed that CAF induced migration NSCLC A549 SPCA-1 this three-dimensional microdevice, confirmed using traditional Transwell system. Furthermore, GRP78 expression facilitate invasion, whereas knockdown blocked capacity. In conclusion, these indicated CAFs might up-regulation microdevice robust platform assess interaction environment study.
