[Polydatin induces human cervical cancer cell apoptosis via PI3K/AKT/mTOR signaling pathway].

ABSTRACT
To observe the effect of polydatin on proliferation and apoptosis cervical cancer HeLa cells explore its possible mechanism. The growth inhibitory was detected with MTT assay. After were treated different concentrations (50, 100, 150 μmol•L⁻¹) polydatin, assay used to detect cells; Acridine orange/ethidium bromide staining for morphological changes in apoptotic cells; Annexin/propidium iodide applied cell rate. In addition, flow cytometry employed analyze cycle distribution; RT-PCR Western blot PI3K, AKT, mTOR, P70S6K mRNA protein expression levels. results showed that significantly inhibited a dose-dependent manner. Polydatin can cause S phase arrest cells, promote decrease levels mTOR P70S6K. It indicated could inhibit induce mechanism may be associated inhibiting PI3K/AKT/mTOR signaling pathway suppressing downstream gene expression.
