Transcriptome analysis and validation of key genes involved in biosynthesis of iridoids in Gentiana lhassica

ABSTRACT
As the main chemical constituents, iridoids are widely distributed within Gentiana, Gentianaceae, with promising bioactivities. Based on previous work, transcriptome of G. lhassica, an original plant Tibetan herb Jieji Nabao, was sequenced and analyzed in this study, databases roots, stems, leaves, flowers were constructed so as to explore unigenes that may encode key enzymes biosynthetic pathway iridoids. Then, qRT-PCR used validate relative expression levels 11 genes named AACT, DXS, MCS, HDS, IDI, GPPS, GES, G10H, 7-DLNGT, 7-DLGT, SLS flowers. Also, total contents gentiopicroside loganic acid determined by HPLC, respectively. The results follows:(1)a 76 486 average length 852 bp obtained;(2)335 involved 19 stan-dard secondary metabolism pathways KEGG database, phenylpropanoid biosynthesis having maximum number(75 unigenes), no isoflavone annotated;(3)171 participatedin 27 encoding iridoids, 1-deoxy-D-xylulose-5-phosphate reductoisomerase(DXR) gene highly expressed;(4)qRT-PCR approximately consistent RNA-Seq data higher aboveground parts(stem, leaf, flower) than underground part(root);(5)the part(root), difference significant. This study provides basic scientific for accurate species identification, evaluation germplasm resources, research pro-duct accumulation medicinal plants protection endangered alpine species.
