[Material basis of toad oil and its pharmacodynamic effect in a mouse model of atopic dermatitis].

ABSTRACT
This study aims to comprehensively analyze the material basis of toad visceral oil(hereafter referred as oil), and explore pharmacological effect oil on atopic dermatitis(AD). Ultra-high performance liquid chromatography-linear ion trap/orbitrap high-resolution mass spectrometry(UHPLC-LTQ-Orbitrap-MS) gas chromatography-mass spectrometry(GC-MS) were employed identify chemical components in oil. The animal model AD was prepared by hapten stimulation method. modeled animals respectively administrated with positive drug(0.1% hydrocortisone butyrate cream) low-and high-doses(1%, 10%) gavage. evaluated score, ear swelling rate, spleen index, pathological section results indicators. A total 99 identified UHPLC-LTQ-Orbitrap-MS, including 14 bufadienolides, 7 fatty acids, 6 alkaloids, 10 ketones, 18 amides, other compounds. After methylation samples, a 20 compounds GC-MS. Compared group, high-dose groups showed declined alleviated skin lesions, reduced infiltrating mast cells. analyzes composition clarifies Meanwhile, this proves that has good therapeutic is reserve resource traditional Chinese medicine for external use treatment AD.
