ABSTRACT
Plaque psoriasis is a common, long-lasting illness that affects the immune system and causes significant negative impacts on patient's physical health, well-being, ability to work effectively. Deucravacitinib (DEU) first oral medication used in treatment of plaque psoriasis, chronic skin condition red, scaly patches skin. DEU type called an Janus kinase (JAK) inhibitor, which works by blocking specific enzymes play role inflammation response associated with psoriasis. Therefore, quick, easy, novel, reliable, sensitive, straightforward Liquid Chromatography-Tandem Mass Spectrometry (LC-MS/MS) approach was analyse plasma samples. The LC-MS/MS method for determination human based using trimethoprim as internal standard (IS). separation IS carried out via liquid-liquid extraction (LLE). isolated substances were then subjected chromatographic ACE-C18 column (4.6x100 mm, 5 µm). mobile phase employed consisted methanol solution 2 mM ammonium formate (80:20 v/v, respectively). flow rate set at 0.9 mL min-1. creative strategy performed running ABSCIEX API 4000 mass spectrometer electron spray ionization source Multiple Reaction Monitoring (MRM) modes. ion transitions m/z 426.3 358.2 quantitation, while 291.1 261.1 quantitation. accuracy, precision, linearity, recovery, selectivity deemed acceptable when validated concentration range between 0.500 601.050 ng/mL, utilizing weighting factor 1/x2.
