Whole Yeast Vaccine Displaying ZIKV B and T Cell Epitopes Induces Humoral and Cellular Immune Responses in Murine Model

ABSTRACT
Improving antigen presentation is crucial for the success of immunization strategies. Yeasts are classically used as biofactories to produce recombinant proteins and efficient vehicles delivery vaccine antigens, besides present adjuvants properties. Despite absence epidemic outbreaks, several approaches continue be developed Zika virus infection. These prophylactic strategies fundamental given severity clinical manifestations, mainly due viral neurotropism. The study aimed evaluate in vivo immune response induced by P. pastoris strains displaying epitopes Envelope (ENV) NS1 ZIKV proteins. Intramuscular with heat-attenuated yeast enhanced secretion IL-6, TNF-α, IFN-γ, activation CD4+ CD8+ T cells, BALB/c mice. ENV a more robust response, increasing immunoglobulin production, especially IgG isotypes. Both proposed vaccines showed potential induce responses without adverse effects, confirming safety administering vehicle. Here we demonstrated, first time, evaluation against based on multiepitope construct, using vehicle, reinforcing applicability whole cell vaccine.
