ABSTRACT
Abstract Background Malaria is a life-threatening infectious disease particularly due to Plasmodium falciparum (P.falciparum ). Histidine-Rich Protein 2 (PfHRP-2) and specific Lactate Dehydrogenase (PfPLDH) based rapid diagnostic test are commonly used for malaria diagnosis in endemic countries where microscopic examination scarce. However, there limited information on the performance of RDT rural semi urban area Assosa zone, Northwest Ethiopia. Thus, aim this study determine PfHRP2 PfPLDH against microscopy as reference method. Methods Health-facility cross-sectional design was conducted Ethiopia from November December 2018. A total four hundred six malaria-suspected participants attending Bambasi, Sherkole, Kurmuk health-centers were tested. Finger-prick blood samples collected film preparation, RDTs molecular diagnosis. Statistical analyses performed using SPSS version 20. Results Of participants, 26.4% (107/406) microscope confirmed P. positive. Using RDT, 30.3% (123/406) 24.1% (98/406) positive , respectively. The sensitivity 96% 89%, respectively, microscope. corresponding specificity rates 93% 99%, Similarly, predictive value (PPV) negative 84%, 97%, 99% 96%, There an agreement between (PfPLDH PfHRP2) method with kappa 0.86 0.90, Compared qPCR, (93%) (98%) high, though respective PfHRP2(77%) RDT(70%) low. Conclusion showed reasonable detecting infections. Hence, currently national kit can be continue certain areas Continuous monitoring PfHRP-2 associated false results important consider alternative like support control elimination
