Restoration of spermatogenesis in azoospermic mice by bone marrow mesenchymal stromal/stem cells conditioned medium

ABSTRACT
Abstract Background One of the main cause male infertility is non-obstructive azoospermia, which not manageable medically. The first aim current research was to show effect extracellular vesicle-contained conditioned media (CM) instead mesenchymal stromal/stem cells (MSCs) for treatment azoospermia. In next step, we aimed study differentiation potential MSCs into spermatocytes after injection in mice seminiferous tubules. This has provided an applied busulfan-induced azoospermia using adipose tissue-derived (AT-MSCs) and bone marrow-derived (BM-MSCs) marrow CM (BMCM) animal models. Methods Thirty adult Balb/C (30±5 g) two female eGFP +/+ were used design experimental groups culture stem cells, respectively. Then, six including intact control, AT-MSC therapy, BM-MSC BMCM spontaneous healing considered. All except control induced by injecting doses busulfan (10 mg/kg) with 21 days’ interval. Testes all removed studied through histomorphometry flow cytometry analysis 60 days treatment. Results Histomorphometry evaluation testes showed normal morphology most tubules therapy as well successful recovery spermatogenesis, but spermatogenesis observed group. It worth notable that results group more favorable than other groups. Conclusions AT-MSC, can be strongly suggested candidates
