ABSTRACT
Metallothionein (MT) plays a central role in cellular defense against heavy metals and oxidative stress. Since the induction of MT requires activation metal response element (MRE)-binding transcription factor-1 (MTF-1) by binding zinc ions, inorganic is regarded as typical inducer. However, previous report, we showed that could not induce vascular endothelial cells. While it suggested presents mechanisms different from those other cell types, these remain unclear. In this study, investigated whether expression involves Nrf2-ARE pathway using copper(II) bis(diethyldithiocarbamate), termed Cu10, culture system bovine aortic Cu10 induced MT-1/2 protein increased mRNAs for MT-1A, MT-1E, MT-2, isoforms expressed activated only MTF-1-MRE, but also pathway. MTF-1 knockdown resulted repression Cu10-induced MT-1 -2 expression. was down-regulated Nrf2 knockdown. MT-2 affected These results suggest up-regulated well MTF-1-MRE Moreover, regulation differ MT-2. Specifically, present data support hypothesis participates biological system, while mainly regulates intracellular metabolism.
