ABSTRACT
C RYPTOSPORIDIOSIS is a significant disease that causes diarrhea in humans and animals with relatively high morbidity mortality.The present study aimed to adopt purified potent antigen for the accurate diagnosis of cryptosporidiosis.A total 278 animal hosts (60 newborn calves 218 buffaloes) were used current study.Sixty fecal samples collected from new-born aged less than one month raised Beni-Suef Qalyubia governorates.The examined under microscope after modified Ziehl-Neelsen staining, Cryptosporidium oocysts isolated naturally infected calves.These mice experimental infection.The oocyst coproantigen prepared feces.The diagnostic efficacy two antigens was evaluated using an Enzyme Linked Immunosorbent Assay (ELISA) experimentally sera.The crude proved have higher potential coproantigen, so, it chosen purification Cyanogen Bromide-activated Sepharose-4B affinity chromatography coupled rabbit hyperimmune serum against antigen.The fraction its ELISA.The resulting 6733 fold increase binding activity compared antigen.Characterization performed sodium dodecyl sulfate polyacrylamide gel electrophoresis (SDS-PAGE), western blot, amino acid analysis.SDS-PAGE clarified contained three polypeptides 94.7, 65, 50 kDa, which identified as immune-reactive components blot analysis.The exhibited 17 acids rich tyrosine, alanine, phenylalanine.The effectively detected antibodies sera buffalo sensitivity 94.4% 95.24 %, specificity 100% 93.33%, respectively.The succeeded cryptosporidiosis 182 random buffaloes incidence 57.14 %.In conclusion, might be good candidate seroepidemiological surveillance.
