ABSTRACT
The human metabolome is influenced by various intrinsic and extrinsic factors. A precondition to identify such biomarkers the comprehensive understanding of composition variability healthy humans. Sample handling aspects have an important impact on metabolome; therefore, it crucial for any metabolomics study standardize protocols sample collection, preanalytical handling, storage, analytics keep nonbiological as low possible.The main objective KarMeN analyze in blood urine targeted untargeted metabolite profiling (gas chromatography-mass spectrometry [GC-MS], GC×GC-MS, liquid [LC-MS/MS], and(1)H nuclear magnetic resonance [NMR] spectroscopy) determine sex, age, body composition, diet, physical activity profiles women men. Here, we report outline protocol with special regard all that should be considered studies applying metabolomics.Healthy men women, aged 18 years or older, were recruited. In addition a number anthropometric (height, weight, mass index, waist circumference, composition), clinical (blood pressure, electrocardiogram, chemistry) functional parameters (lung function, arterial stiffness), resting metabolic rate, activity, fitness, dietary intake assessed, 24-hour urine, fasting spot plasma samples collected. Standard operating procedures established steps design. Using different analytical techniques (LC-MS, GC×GC-MS,(1)H NMR spectroscopy), determined. Data will analyzed using univariate multivariate well predictive modeling methods.The project was funded 2011 enrollment carried out between March 2012 July 2013. total 301 volunteers eligible participate study.
