ABSTRACT
To develop a mouse orthodontic organ culture model and examine early-induced changes in osteoblast differentiation markers within the periodontal ligament (PDL) alveolar bone.Mandibles from 4- to 12-week-old transgenic mice were dissected hemisected. A conventional superelastic spring (25 grams) was bonded incisor first molar on one side of mandible; other served as control. Dissected mandibles cultured for 6 hours then histologically analyzed proliferation (BrdU immunostaining) fluorescent protein expression. Additionally, an vivo using same methods applied 3.6 Col1-GFP mice.In vitro, after loading, significant increase noted 3.6Col1-GFP- BSP-GFP-positive cells tension PDL compared with unloaded controls. On compression side, decrease positive 3.6Col1-GFP observed In vivo, tendencies found.This novel vitro mandibular tooth movement coupled technology provides powerful tool delineating initial cellular molecular events movement.
