ABSTRACT
OBJECTIVE To investigate the longitudinal development of neurofilament light chain (NfL) levels in type 2 diabetes with and without diabetic polyneuropathy (+/−DPN) to explore predictive potential NfL as a biomarker for DPN. RESEARCH DESIGN AND METHODS We performed retrospective case-control analysis data from 178 participants Anglo-Danish-Dutch Study Intensive Treatment People Screen-Detected Diabetes Primary Care-Denmark (ADDITION-Denmark) cohort people screen-detected diabetes. Biobank samples acquired at ADDITION-Denmark 5- 10-year follow-ups were analyzed serum (s-NfL) using single-molecule array, results compared established reference material obtain z-scores. DPN was diagnosed according Toronto criteria confirmed follow-up. RESULTS s-NfL increased over time +DPN (N = 39) −DPN 139) above normal age-induced increase. Longitudinal change greater than (17.4% [95% CI 4.3; 32.2] or 0.31 SD 0.03; 0.60] higher z-score increase −DPN). 5-year follow-up positively associated nerve conduction studies (P 0.02 <0.001), but not risk. Areas under curve (AUCs) inferior AUCs Michigan Neuropathy Screening Instrument questionnaire score vibration detection thresholds. Higher yearly risk (odds ratio 1.36 1.08; 1.71] per 1 ng/L/year). CONCLUSIONS Our findings suggest that preceding trajectories differ slightly between those imply possible value
