ABSTRACT
Nitric oxide (NO) is known to be a neuromodulator with dual proconvulsive and anticonvulsive action.Valeriana officinalis (VAL) was previously believed antiepileptic, but today as sedative sleep regulator.Seizures may associated abnormal electrocardiographic changes cardiac dysfunction arising from epilepsy related neuronal nitric (nNO).This study aimed investigate the effects of synthase (nNOS) inhibitor 7-Nitroindazole (7-NI) VAL on seizure behaviours parameters in pentylentetrazole (PTZ)-kindled model.Wistar rats were randomised into saline control, PTZ-kindled, 7-NI, VAL+PTZ, 7-NI+PTZ VAL+7-NI+PTZ groups.Latency, stage, frequency seizures, blood pressure (BP), heart rate (HR) corrected QT (QTc) values evaluated.Frequency stage BP HR increased, while latency decreased QTc prolonged PTZ-kindled group.7-NI had no variables under normal conditions, ameliorated seizures.7-NI treatment also resulted reduction increased observed rats.Considering these results, prolongation used predictor for recurrent exhibited different seizures ECG variables.7-NI shows potential an anticonvulsant drug agent epileptic patients dysfunctions those additional studies including in-vivo experiments are essential.
