ABSTRACT
99mTc-annexin-V imaging has been proved to be feasible detect phosphatidylserine, which externalizes on the outer cell membrane early in process of apoptosis. To determine whether postconditioning suppresses myocardial damage or apoptosis, we evaluated intensity and distribution uptake after a rat model ischemia reperfusion compared effect that ischemic preconditioning pretreatment with caspase inhibitor. In control rats (n = 13), thoracotomy left coronary artery was occluded for 20 min followed by 30 90 injection (80-150 MBq). One hour later, verify area at risk, 201Tl (0.74 MBq) injected intravenously just beyond reocclusion, were sacrificed 1 later. In groups various interventions, 11) performed reperfusion, inhibitor treatment before ischemia. Dual-tracer autoradiography assess risk.In all rats, intense observed risk (uptake ratios 4.15 +/- 1.89 3.70 1.41, respectively). Postconditioning suppressed 2.09 0.56, P < 0.05, 1.88 0.69, Preconditioning also 1.17 0.29, 0.005, 1.33 0.74, 0.01, respectively), as did 2.08 0.50, 1.27 0.24, percentage cells positive deoxyuride-5'-triphosphate biotin nick end labeling histologic changes degeneration infiltrations markedly. These data indicate may way monitor injury its response novel therapeutic interventions including postconditioning, preconditioning, antiapoptotic therapy.
