ABSTRACT
Buprenorphine is commonly used to control postoperative pain in rodents. Short-acting formulations of buprenorphine (bup-HCl) require frequent handling and restraint animals for appropriate dosing, which can be stressful confound research outcomes. Ethiqa XR (bup-ER) an FDA-indexed extended-release formulation that alternative bup-HCl mice rats. In the current study, we first evaluated pharmacokinetics bup-ER male C57BL/6J by sampling blood at 10 time points, ranging from 30 min 72 h after administration ( n = 3 per point). Average plasma concentrations fell below therapeutic levels 48 administration. We also safety when administered prior surgery combination with common anesthetics efficacy mouse laparotomy. Anesthetic was studied measuring respiratory rate, rectal temperature, recovery groups 8) given bup-HCl, bup-ER, or saline isoflurane ketamine-xylazine anesthesia. No differences were seen between analgesic treatment either general anesthetics. To evaluate efficacy, 10) randomly allocated receive (3.25 mg/kg) once presurgically, (0.1 presurgically then every 8 h, before surgery. Mice underwent a sham laparotomy assessed based on changes weight, cageside ethogram, nesting consolidation test, rearing frequency, nociception von Frey testing 6, 12, 24, 48, Cageside showed significant bup-ER-treated controls early period. effects daily weights scores. This study demonstrates safely provides analgesia up pharmacokinetic behavioral data.
