ABSTRACT
Lanthanum, a rare earth element, has been used to decrease serum phosphorus level in patients with chronic renal disease and hyperphosphatemia. We aimed observe the effect mechanism of two doses lanthanum acetate (375 750 mg/kg/day) on vascular calcification induced by vitamin D3 plus nicotine treatment rats for 4 weeks. As compared control rats, showed widespread calcified nodules irregular elastic fibers aorta von Kossa calcium staining increased aortic contents, alkaline phosphatase (ALP) activity bone-related protein expressions osteopontin (OPN) type III sodium dependent phosphate cotransporter Pit-1 (Pit-1). After either dose acetate, degree decreased aortas. Lanthanum at mg/kg/day was more effective than 375 lessening significantly reducing plasma level, x product ALP activity, 30.3%, 28.6%, 68.6%, respectively; contents 48%, 53.1%, 63.5% (all P < 0.01), mRNA OPN Pit-1, 55.8% (P 0.01) 38.8% 0.05) 37.2% 27.2% (both increasing carboxylated matrix Gla-protein (MGP) expression 33.7% 0.05), as treated alone. could effectively inhibit pathogenesis calcification.
