ABSTRACT
MSC senescence is considered a contributing factor in aging-related diseases. We investigated the influence of inflammatory microenvironment on bone marrow mesenchymal stem cells (BMSCs) under aging conditions and underlying mechanism to provide new ideas for cell therapy age-related osteoporosis. The BMSCs were cultured until passage 3 (P3) (young group) 10 (P10) (aging vitro. supernatant was collected as conditioned medium (CM). young CM P3 or P10 cells. effects from different groups stemness examined. A Quantibody® mouse inflammation array serum extracts (aged 8 weeks) old 78 mice performed, differentially expressed factors screened out. discovered that senescent MSCs changed physiology BMSCs. Systemic microenvironments with age mice. In particular, pro-inflammatory cytokine IL-6 increased, anti-inflammatory IL-10 decreased. by GO KEGG analyses, there change JAK-STAT signaling pathway, which closely related IL-10. Collectively, our results demonstrated has significant effect biological functions Targeted reversal this environment may strategy treat skeletal
