ABSTRACT
Ginsenoside Rg3 is a steroidal saponin isolated from Panax ginseng. Previous studies have shown that treatment downregulates the activity of rapamycin complex 1 (mTORC1) and inhibits growth cancer cells. However, inhibitory effect on cells associated with high concentrations are difficult to achieve in vivo. The human cervix adenocarcinoma HeLa were treated Rg3. protein levels AMP-activated kinase alpha (AMPKα), B(Akt), ribosomal S6 protein(S6), Erk determined by immunoblotting analyses. We used fluorescent probe detect reactive oxygen species (ROS) production living consumption rate was examined Seahorse. Extracellular Flux Analyzer. content ATP measured ATPlite kit Mitotracker applied mitochondria. showed at lower concentrations, activates mTORC1 independent AKT AMPK. promotes mitochondrial biogenesis function, increases mitochondria ATP. This contrast Rg3, which cell growth. These findings demonstrate pro-growth acts through suggest dose-dependent tumor
