ABSTRACT
Background: Elevated expression of Copine-1 (CPNE1) has been proved in various cancers; however, the underlying mechanisms by which it affects clear cell renal carcinoma (ccRCC) are unclear. Methods: In this study, we applied multiple bioinformatic databases to analyze and clinical significance CPNE1 ccRCC. Co-expression analysis functional enrichment were investigated LinkedOmics, cBioPortal Metascape. The relationships between tumor immunology explored using ESTIMATE CIBERSORT method. vitro experiments, CCK-8, wound healing, transwell assays western blotting conducted investigate effects gain- or loss-of-function ccRCC cells. Results: was notably elevated tissues cells, significantly correlated with grade, invasion range, stage distant metastasis. Kaplan-Meier Cox regression displayed that an independent prognostic factor for patients. Functional revealed its co-expressed genes mainly regulated cancer-related immune-related pathways. Immune correlation showed related immune estimate scores. positively higher infiltrations such as CD8+ T plasma cells regulatory exhibited lower neutrophils. Meanwhile, characterized high infiltration levels, increased levels exhaustion markers (CTLA4, PDCD1 LAG3) worse response immunotherapy. studies demonstrated promoted proliferation, migration through EGFR/STAT3 pathway. Conclusion: is a reliable predictor prognosis promotes proliferation activating signaling. Moreover, correlates
