ABSTRACT
Introduction: Porphyromonas gingivalis and Enterococcus faecalis promote the development of pulpitis periapical periodontitis. These bacteria are difficult to eliminate from root canal systems, leading persistent infection poor treatment outcomes. We explored response human dental pulp stem cells (hDPSCs) bacterial invasion mechanisms underlying impact residual on regeneration. Methods: Single-cell sequencing was used categorize hDPSCs into clusters based their P. E. . depicted a single-cell transcriptome atlas stimulated by or Results: The most differentially expressed genes in Pg samples were THBS1 , COL1A2 CRIM1 STC1 which related matrix formation mineralization, HILPDA PLIN2 cellular hypoxia. A cell cluster characterized high expression levels PTGS2 increased after stimulation. Further signaling pathway analysis showed that prevented regulating TGF-β/SMAD, NF-κB, MAPK/ERK pathways. Differentiation potency pseudotime trajectory analyses infected undergo multidirectional differentiation, particularly mineralization-related lineage. Furthermore, can create hypoxia environment effect differentiation. Ef CCL2 is leukocyte chemotaxis, ACTA2 actin. There an proportion similar myofibroblasts exhibited significant expression. presence promoted differentiation fibroblast-like cells, highlights role tissue repair. Discussion: do not maintain status They differentiate identified mechanism Our results will improve understanding pathogenesis have adverse effects outcomes regenerative endodontic treatment.
