Transcriptome and proteomic analysis of mpox virus F3L-expressing cells

ABSTRACT
Background Monkeypox or mpox virus (mpox) is a double-stranded DNA that poses significant threat to global public health security. The F3 protein, encoded by mpox, an apoenzyme believed possess RNA-binding domain (dsRBD). However, limited research has been conducted on its function. In this study, we present data the transcriptomics and proteomics of F3L-transfected HEK293T cells, aiming enhance our comprehension F3L. Methods gene expression profiles pCAGGS-HA-F3L transfected cells were analyzed using RNA-seq. Proteomics was used identify study proteins interact with Real-time PCR detect mRNA levels several differentially expressed genes (DEGs) in (or Vero cells) after protein. Results A total 14,822 obtained RNA-Seq 1,672 DEGs identified, including 1,156 up-regulated 516 down-regulated genes. 27 cellular interacting identified liquid chromatography-tandem mass spectrometry (LC-MS/MS), 19 large differences abundance ratios considered be candidate proteins. Gene Ontology (GO) Kyoto Encyclopedia Genes Genomes (KEGG) enrichment analyses showed significantly enriched immune-related pathways, type I interferon signaling pathway, response virus, RIG-I-like receptor NOD-like etc. Moreover, some selected further confirmed real-time results consistent transcriptome data. show are mainly related RNA splicing protein translation. Conclusions Our analysis transcriptomic proteomic (1) F3L up-regulates transcript key innate immune such as RIGI, MDA5, IRF5, IRF7, IRF9, ISG15, IFNA14 , elicits broad spectrum antiviral responses host. also increases FOS JNK while decreasing TNFR2, these factors may ultimately induce apoptosis.
