ABSTRACT
The most common type of pituitary neoplasms is benign adenoma (PA). Clinically significant PAs affect around 0.1% the population. Currently, there no established human PA cell culture available and when tumour cells are cultured they form two distinct types depending on culturing conditions either free-floating aggregates also known as pituispheres or adhering to surface plates displaying mesenchymal stem-like properties. aim this study was trace origin sphere-forming adherent cultures characterize potential use these surgery derived lines model. We carried out a paired-end exome sequencing patients' germline DNA using Illumina NextSeq followed by characterization corresponding cultures. Variation analysis revealed low amount somatic mutations (mean = 5.2, range 3 - 7) in exomes PAs. Somatic primary material can be detected respective pituispheres, but not cells. For first time, we show that genome represents while stem from tissue do contain characteristic their genome, therefore, likely representing normal surrounding tissues. This finding indicates used model cells, combination techniques NGS needs employed adjust for disability propagate spheres conditions.
