ABSTRACT
Background Laparoscopic sleeve gastrectomy (LSG) is a sustainable technique that effectively treats morbid obesity. However, the molecular mechanisms underlying improvement of metabolic health following this process warrants more investigation. This study investigates LSG-related molecules and uses bulk RNA-sequencing high-throughput analysis to unravel their regulatory mechanisms. Methods Peripheral blood mononuclear cells (PBMC) were collected from ten obese patients with BMI ≥ 32.5 kg/m 2 in Department General Surgery Kunming First People’s Hospital. After LSG, followed up for one month, samples retaken. Blood before after LSG RNA-Seq data analyzed study. LSG-associated gene expression was detected by weighted coexpression network (WGCNA) differential analysis. Subsequently, essential signature genes identified using logistic least absolute shrinkage selection operator (LASSO) support vector machine-recursive feature elimination (SVM-RFE) algorithms. Gene Ontology (GO), Kyoto Encyclopedia Genes Genomes (KEGG), single-sample set enrichment (ssGSEA) utilized reveal potential functions target genes. Furthermore, Pearson correlation leptin lipocalin also explored. Finally, we constructed robust endogenous RNA (ceRNA) based on miRWalk starBase databases. Results We 18 overlapping 91 hub genes, 165 differentially expressed mRNAs (DE-mRNA), which revealed be significantly associated immune cells, response, inflammatory lipid storage, localization upon functional Three IRF1 , NFKBIA YRDC LASSO SVM-REF The regression model three highlighted how robustly they discriminated between samples. ssGSEA indicated these involved metabolism degradation pathways. Moreover, levels reduced undergoing negatively correlated leptin. long non-coding (lncRNA) ATP2B1-AS1 regulated competitively binding six microRNAs (miRNAs), hsa-miR-6509-5p, hsa-miR-330-5P, hsa-miR-154-5P, hsa-miR-145-5P, hsa-miR4726-5P hsa-miR-134-5P.
