ABSTRACT
Introduction In the Dongting water system, Carassius auratus (Crucian carp) complex is characterized by coexistence of diploid forms (2n=100, 2nCC) and polyploidy forms. The (2nCC) triploid C.auratus (3n=150, 3nCC) had same fertility levels, reaching sexual maturity at one year. Methods nucleotide sequence, gene expression, methylation, immunofluorescence gonadotropin releasing hormone 2( Gnrh2 ), Gonadotropin beta( Gthβ Gonadotropin-releasing receptor( Gthr ) genes pivotal hypothalamic-pituitary-gonadal (HPG) axis were analyzed. Results analysis results indicated that , follicle-stimulating Fshr Lethal hybrid rescue( Lhr increased copy number distinct structural differentiation in 3nCC compared to 2nCC. transcript levels HPG higher than 2nCC (P<0.05), which could promote production secretion sex steroid hormones conducive gonadal development 3nCC. Meanwhile, DNA methylation promoter regions lower These suggested region a potential regulatory effect on expression after triploidization. Immunofluorescence showed localization Fshβ, Lhβ between remained unchanged, ensuring normal these corresponding sites Discussion Relevant research provide cell molecular biology evidence for reproductive activities such as gonad gamete maturation C. contribute further understanding genetic basis restoration .
