ABSTRACT
Introduction The longitudinal section cell morphology of Chinese fir roots was studied to better understand the relationship between root structure and function. Materials In this study, adjusted microwave paraffin method selected two sample transparency methods were used process combined with laser scanning confocal microscopy (LSCM) technique, can be clearly observed in a short time. At same time, observation effect LSCM image thick ordinary optical imaging thin analyzed compared. Results Discussion results showed that: (1) There apparent differences sections obtained using various treatment methods. Under LSCM, thickness 20 μm generated by technique displayed complete clear cap, meristem zone, elongation zone. overall good; 0.42–1.01, 0.64–1.57, 0.95–2.71 mm, respectively. arrangement maturation zone cells more regular. (2) Compared sections, made shortened time obtain high-quality ensure effect. Therefore, adopting cutting approach produce thicker under allows for rapid roots. current study provides efficient operation procedure microscopic technology roots, which is not only beneficial reveal function from point view but also technical reference anatomical other organs plant similar structural characteristics.
